No, boards marks are not calculated in your rank. It is on the basis of marks scored in your MHT CET exam.
You have to fulfill the minimum eligibility criteria of the exam that is you should have 50% aggregate marks.

Below are the some of the changes in 2018
1- The difficulty level of MHT CET 2018 will be equivalent to JEE Main
2- The syllabus of MHT CET 2018 will be based on Maharashtra State Board of Secondary and Higher Education
Around 20% weigtage will be given to Class XI curriculum while Class XII curriculum will carry remaining 80% weightage
3- There will be no negative marking

No, 12th percentage is not required in MHTCET as 50% is mandatory to apply for the same as an eligibility criteria only. Also, to get admission in MHTCET only its score is important as it does not give weightage to the 12th score. And to get admission in top colleges of Maharashtra like COEP, VJTI etc, in top branches more than 170 would be required.
