Yes, diploma holders of engineering branch are eligible to apply for an entrabce exam for B.tech courses.
Some universities take the entrance exams to get lateral entry admission in second year B.tech courses, while some university take admission on the basis of the merit marks. After successfully completing the 3 year diploma course you are eligible to take admission to any B.tech courses from second year. This is called lateral entry admission.

Yes definitely, any diploma holder can pursue B.Tech degree in the same domain as that of diploma.
Diploma is considered as equivalent to the 12th class exam, provided done by government recognized institute.
Also, as you have mentioned that you have completed 3 years diploma, then you can apply for 2nd year in B.tech directly. This is called Lateral entry and you can write an entrance exam for same, known as LEET (Lateral Entry Entrance Exam).
If you have completed the diploma in civil engineering then you can apply for BTech in Civil Engineering in various colleges.
