Yes there is a separate entrance exam for admission in DU. You can apply for it by filling out an online form on its official website.
However, entrance exams are held only for a few select courses like Journalism and Business Management.
For most Arts, science and commerce courses (eg- Physics Hons, English Hons, B.Com (H), Bsc in Zoology) etc, admissions are merit based. It depends on the candidate's performance in Higher secondary (class 12) examination from their respective boards.

No, entrances are not conducted by the University of Delhi, for admissions to the any College of the university.
The admissions to almost all courses offered at the undergraduate level are merit-based.
The university conducts entrances only for admissions to bachelor's in management studies , bachelor's in business economics and bfia courses.
The entrance test is called the Delhi university joint entrance examination.
