I have completed class 12 with PCB from CBSE in 2020. But now I want to pursue class 12 again but with PCMB to get admission into IISc or IISER. Is it possible? please help me.

Hi Dear,
YOu can get into Indian Institute of Science (IISc) and Indian Institute of Science Education and Research even after completion of your 10+2 from physics, Chemistry, and Biology stream. You do not need to repeat your 12th class for this purpose. You can either enroll into undergraduate research programmes now or you can also prepare for integrated PhD and MSC course from IISC and IISER after completion of your graduation the same field.

But on the website of iisc, it is written that eligible students should complete their 10+2 with physics, Chemistry and Mathematics, in addition they can have biology as an additional subject.

For postgraduate programs mathematics is not mandatory.

Yes you are eligible. You need to have either mathematics or Biology in 10+2.
