I have got a rank of 25,949 OBC rank in JEE Main 2020. I'm having home state quota of Himachal pradesh. Can i get CSE 4 year programe at NIT Hamirpur in further rounds as 1st round closing rank was 25431??

First of all congratulations as got have secured 25949 obc rank in jee mains 2020.
As your home state is himachal pradesh, then you can easily get nit hamirapur via your home state quota. As you said that for first round the cutoff closing rank is 25431 for cse in nit hamirpur, then there is a high chances that you can get cse branch before 5th round counselling.
Also there is a high chance that you can get cse in nit hamirpur as per seeing last year cutoff trends. Last year the cutoff closing rank for obc candidates for cse Branch in nit hamirpur was - 28329.
So, there is a high chance that you can get cse in nit hamirpur in next rounds.

Actually, in second round too, I didn't get that branch. So, I'm worried now.

there is a chance that you can easily get cse branch as per seeing last year cutoff trends. but last year in 2nd round the cutoff closing rank was - 28329 for cse. and this cutoff closing but remained same tool 7th round counseling. aap it's better that you do not take any risk. use the float option.
