In first cap round I didn't get any college can I directly fill second option form cap round 2

As you've not mentioned about which exam you're talking about, so please clear it a little bit. Although as I can assume that CAP rounds are in MHT CET so I'll give my answer regarding that.
Yes, you can directly fill your options for 2nd cap round given that you satisfy the all the criteria of the conductin committee.
