NO, Mathematics Paper which is a part of session 1 is only needed if you are applying for M.Sc. in mathematics

Mathematics as a part of syllabus is also included in the Physics Test (Session 1) and Mathematical Statistics Test (Session 2)

The IIT-JAM syllabi depends on the course for which you are applying.
