Hello aspirant,
I hope that you are doing absolutely great.
With reference to your above mentioned query, I would like to tell you that yes, you can obviously apply for and can give your JEE mains and advanced examination after completing your 2nd year of diploma. JEE mains and advanced exams is an important entrance exam that every candidates need to give in order to get admission in top NITs or top IITs.
Due to high course fees, students are unable to enroll themselves in Bachelor of Engineering or Bachelor of technology. So the students enroll themselves in a diploma in engineering course in order to get direct admission into the 2nd year of B.tech and B.E programme by a lateral scheme. Diploma in engineering acts as a back door entry into the B.tech/B.E reducing the overall fee for students taking up a diploma course.

I hope that this will help.
Yes, absolutely possible.
You'll need to qualify JEE Mains and JEE Advanced to get admission for your first year of four year degree course. Many state and private colleges are admitting the diploma holders directly to the second year, but neither IITs nor NITs are assisting this. After completing Diploma in Engineering graduates can apply for the JEE MAINS exam. You have to strengthen your knowledge in Physics, Chemistry and most importantly in Mathematics. Only passing the JEE MAINS cannot grab your admissions in Engineering colleges. You have to excel in both JEE MAINS and ADVANCED. You will not be considered for lateral entry positions during admissions. You will have to join the first year of the Engineering Program like other candidates, who completed their 12th std.
I would suggest you to take up LEET (Lateral Entry Entrance Exam). This exam is vitally for diploma holders, who desires to get admissions for BE and B.Tech. Primarily, it can be a difficult process to face JEE after diploma. Whereas, this exam will take you through the journey, where you can sharpen your learning curve by taking a deep dive into basics and fundamentals. Subsequently, prepare for GATE exams investing your smart work, genuine effort and sincerity. You can sail through the GATE with the exceptional marks.
All the best!
