In some cases, yes it is mandatory. About every state has rural posting bond except for Delhi.
Delhi is having no rural areas, there are no villages in Delhi, therefore there is not rural bond.
Refer to site : AIPMEE, Under the tab ‘Participating institutions’, you will get all bond related information you need for each and every medical college via neet ug all India quota.
To sum it you do NOT have mbbs rural bonds yet in colleges of following states-Delhi, HP, UP, Bihar, Jharkhand, Punjab, Chandigarh, Haryana, Kerala, West Bengal, Orissa, North eastern states (except AMC, Dibrugarh ), Puducherry, MMC and few colleges in Tamil Nadu.
Central institutes like AIIMS,JIPMER, IMS-BHU, AMU etc. have no service bonds attached.
Apart from these all other states have some kind of rural service bonds varying from one year to five years - Rajasthan, Gujarat, MP, Maharashtra , Goa, Karnataka, Chhattisgarh, Uttarakhand, Andaman & Nicobar.
