Is it necessary to upload signature with black ink pen during filling neet 2020 application form, signature with blue is not valid?

Yes it is necessary to upload your signature with black ink pen while filling NEET 2020 application form. You cannot do it with a blue pen according to their guidelines.
The correction window will open from 15th January to 31st January 2020. You can correct and upload the new signature at that time.
