My brother is 19 at completion of 12th class does age have any negative effect on career( jobs) because he lost 1 year

Not really.There won't be any issue whatsoever regarding his job.Though if he will opt for any other competitive exam where there is age eligibility criteria then he won't be able to appear for the exam.
Though in job sector he wont face any issue.

No definitely not there will not be any side effect even he has one year gap. If he is doing b. Tech then there is generally criteria that one should not have more than 1 or 2 year gap. So he has only one year gap. So take care that there may not be another year gap.
