Hey,
Yes, if you want to take the admission in DU via entrance test basis then you have to give DUET and if you want it on merit basis of 12th marks that is the cutoff list of each college then fill the DU application form.
It depends on you whether you want to take both on either of the two.

No, if you want admission in entrance based colleges and courses then you just have to fill DUET application form.DU application form is mainly for ug courses and is merit based.is Delhi University Entrance Test which is conducted by Delhi University for offering admission in UG, PG, MPhil and PhD programmes of various colleges.
