Whether I have to lock choices manually or it will lock itself in closing time (5pm),after I have completed my choice filling.

if you can lock choices then it will be good as you can check and confirm then but even if you don't lock them the selected choices of yours will automatically get locked after the closing time so it won't be a problem for you but it will better for you if you confirm it and lock it yourself so you don't miss any colleges
