Hello there,
NTA takes 36 to 72 hours for replying to the queries sent by the candidates in their helpline email id. If you have any emergency, then you may try calling them at their helpline number too. Else sit back and relax, they generally respond within 72 hours.

But do they reply back surely?

Yes they do. but that actually depends on the question as for example last year students asked them through emails whether jee main exam will be postponed to which they can't answer privately . So if you have any other questions like registration, fees or admit card, they would surely answer you.
