No, the fresh choice filling will not be available for second round of tamilnadu neet pg 2020 counselling. As According to the counselling guidelines given in the tamil nadu state neet pg counselling
"A candidate who is absent during 1st round of counselling will not be permitted to attend the subsequent rounds of counselling. (i.e. second round and mop-up round)."

I got seat in first round and I joined my college..if I get upgraded in next counselling whether my current seat get canceled?

No, usually on upgrading to round 2 the seat allotted in round one does not cancel. It will cancel when one gets a seat in round 2. The notification for round 2 has not been released by tamil nadu neet pg counselling authority when it will released the procedure for round 2 counselling so wait for the notification. It will clear all your doubts.
